What Is a Pyrogen?

A pyrogen is any substance that causes fever when introduced into the body. Bacterial endotoxin (LPS) is the most common and potent pyrogen. Pyrogen testing is mandatory for all injectable peptide products. Depyrogenation of glassware (250°C dry heat for 30 min) and endotoxin-free water are essential for pyrogen-free peptide preparation.

  • LAL test: Limulus Amebocyte Lysate detects endotoxin specifically. < 0.25 EU/mL for injection water
  • Rabbit pyrogen test: Historic standard. Measures temperature rise after IV injection
