What Is a Peptide Bond?

A peptide bond is a covalent amide bond formed between the alpha-carboxyl group of one amino acid and the alpha-amino group of another, with the elimination of one molecule of water (a condensation reaction). It is the fundamental linkage that holds all peptides and proteins together, and every peptide consists of a chain of amino acids connected by these bonds.

Chemical Properties

The peptide bond has partial double-bond character due to resonance between the carbonyl C=O and the C-N bond. This restricts rotation around the C-N axis, forcing the six atoms of the peptide unit (Cα-CO-NH-Cα) into a planar configuration. The bond is predominantly in the trans configuration (99.9% of cases), with cis bonds occurring primarily before proline residues.

The peptide bond absorbs UV light at 214nm, which is why this wavelength is used for peptide detection in HPLC. This absorption is proportional to the number of peptide bonds, making it a reliable detection method regardless of amino acid composition.

Formation in Synthesis

In solid-phase peptide synthesis, peptide bond formation is driven by activating the carboxyl group with coupling reagents (HBTU, HATU, or DIC/HOBt) to create a reactive intermediate that reacts with the free amino group of the growing chain. Coupling efficiency must exceed 99% per step, as even small losses accumulate exponentially over long sequences.

Stability and Cleavage

Peptide bonds are thermodynamically stable under physiological conditions but are readily cleaved by proteases. The specificity of protease cleavage depends on the amino acids flanking the bond, forming the basis of peptide proteolysis studies and peptide mapping protocols.
