What Does Hygroscopic Mean?

A hygroscopic substance readily absorbs moisture from the atmosphere. Many lyophilized peptides are hygroscopic, especially those with TFA counterions or polar residues. Moisture absorption decreases net peptide content, promotes degradation (hydrolysis, deamidation), and can cause cake collapse during storage.

Management

  • Storage: Desiccated container, -20°C, sealed with parafilm. Equilibrate to RT before opening
  • Weighing: Weigh quickly. Use dry gloves. Return to desiccator immediately
  • Water content: Karl Fischer titration quantifies residual moisture. Target < 3% for stability

Frequently Asked Questions
